733 3D electron microscopy revealed that lamellar granules are secreted on the underside of more matured granular cells

Background: Lamellar granules (LGs) are essential cell organelles secreted extracellularly and contribute to skin barrier function. Our previous 3D electron microscopy (EM) studies revealed that LGs remains in the cytoplasm 3rd layer from surface of epidermal granule layer, SG3, secretion toward begins 2nd SG2, with all 1st SG1. However, details regulatory mechanism still need be fully elucidated. Aim: To clarify secretory regulation LGs. Materials & Methods: Healthy human samples were subjected 2D transmission EM (TEM) 3DEM. For latter, we used serial section scanning (SEM), which uses imaging resin-embedded thin sections on glass slides. We cut 300 100 nm thickness examined them by SEM. Granular cells, nuclei, extracellular segmented, images reconstructed. Results: 2DEM showed when SG2 cells contact multiple SG1 LG does not occur uniformly underside contact. In 2DEM, however, cellular phenomena always correctly reflected a particular cross-section, positional relationship is uncertain. 3DEM if an faced two or more can limited cell, was located closer stratum corneum than other cells. Conclusion: Granule may able sense degree differentiation they powerful tool for unraveling biology.
